Fig. 1. Activation efficiency of human T cells was evaluated by flow cytometry using activation markers CD25 and CD69. Cells were evaluated 24 hours after stimulation and compared. (A) Negative control, (B) ACROBiosystems CD3/CD28 beads (Cat. No. MBS-C001) and (C) competitor products were compared. ACROBiosystems CD3/CD28 beads show superior activation rates compared to competitor’s T cell activation beads.

Fig. 3. Purified human T cells from a single donor were activated using ACROBiosystems CD3/CD28 Beads (Cat. No. MBS-C001) and expanded for 2 weeks in four different, commonly used cell medias supplemented with Human IL-2 (Cat. No. IL2-H5215, 4ng/mL). High cell viabilities and fold expansions were observed, highlighting compatibility with most types of cell media.

Fig. 4. Comparison of CD3+ T cell isolation capabilities of ACROBiosytems CD3/CD28 beads (Cat. No. MBS-C001) against competitor product using human PBMCs. (A) The PBMC population was used as a control, with the unbound cell population remaining in the cell supernatant being evaluated. Isolation was performed using (B) ACROBiosystem CD3/CD28 beads and (C) competitor products (Competitor T) for comparison. Residual CD3+ T cells remaining in the supernatant was evaluated through FACS, revealing a lower percentage of non-isolated CD3+ T cells. (D) Recovery rate of CD3+ T cells were also compared, showing no losses in isolation efficiency with (E) a high level of CD3+ T cell population purity.

Fig. 5. Cell characterization, composition, and PD-1 expression after expansion was performed and compared for both ACROBiosystem CD3/CD28 beads (Cat. No. MBS-C001) and competitor beads (Competitor T). Isolated human CD3+ T cells were cultured for 13 days and analyzed. (A) On day 13, cell composition (naïve T cells, Tn; T memory stem cells, Tscm; central memory T cells, Tcm; effector memory T cells, Tem; terminal effector T cells, Tte) were evaluated using flow cytometry. Expansion performed using ACROBiosystem CD3/CD28 beads resulted in improved percentage of naïve T cells while minimizing Tte cells. (B) PD-1, a surface marker for cell exhaustion, was also evaluated with ACROBiosystems’ beads having lower PD-1 expression and thus, lower cell exhaustion through the cell expansion process.
