背景
组织相容性复合物MHC
免疫系统的核心职能是负责识别自我、排除非我以维持机体的正常生理活动和阻止外源病原体侵袭的能力,此功能在很大程度上是由主要组织相容性复合体MHC(Major histocompatibility complex)分子主导,MHC在人类中也称为HLA。MHC主要分为MHC-I和MHC-II类分子:MHC-Ⅰ类分子几乎分布于所有核细胞表面,重要生理功能是参与向CD8+T(杀伤T细胞)细胞递呈抗原的过程,且CD8+T细胞只能识别与MHC-Ⅰ类分子结合的抗原,多为内源性细胞抗原。MHC-II类分子的分布比较局限,主要表达于B细胞、单核-巨噬细胞和树突状细胞等抗原递呈细胞上,II类分子的功能主要是在免疫应答的始动阶段将经过处理的抗原片段递呈给CD4+T细胞,且多参与外源性抗原的递呈。
TCR与MHC
T 细胞上表达的 T 细胞受体TCR(T Cell Receptor)可特异性识别MHC分子与肽片段的复合物并与其结合,与免疫应答和抗病性能关系密切。如抗原特异性的细胞毒性T细胞(CTL)识别的主要是结合MHC-I类分子的内源性抗原肽。CTL通过TCR识别并结合递呈在靶细胞表面MHC-I类分子肽结合槽内的特异性抗原表位(epitope),在共同刺激信号的协助下活化、增殖,形成表位特异性T细胞克隆,活化的CTL细胞再通过相同的机制与靶细胞结合而发挥生理效应。

No. HL1-HP2E5) and negative control protein respectively. PE signal was used to evaluate the binding activity (QC tested).
