集落刺激因子(CSFs)及其受体
背景
集落刺激因子(Colony stimulating factor, CSFs)是一组在体内外均能强烈刺激骨髓造血干细胞增殖、分化并形成细胞集落的低分子细胞因子。CSFs能与造血干细胞表面的受体蛋白结合,激活下游信号,促进特定种类的血细胞(通常是白细胞,如中性粒细胞)的产生,以应对感染。研究表明,CSFs还可以通过免疫依赖和非免疫依赖的途径在多种恶性肿瘤中表达上调并促进肿瘤细胞的生长、侵袭 和迁移。另外,CSFs还被发现可以通过调节肿瘤微环境,在多种恶性肿瘤中发挥抗肿瘤效应。因此,CSFs和CSFRs是血细胞发生必不可少的刺激因子且在多种实体肿瘤中具有潜在预后价值。

药物靶点

M_CSF系列靶点蛋白

巨噬细胞集落刺激因子(M-CSF)与其受体(M-CSFR)结合可促进单核细胞存活、单核细胞向巨噬细胞的转化和巨噬细胞增殖。它还能启动并增强巨噬细胞对肿瘤细胞和微生物的杀伤作用,调节巨噬细胞释放细胞因子和其他炎症调节剂,并刺激胞饮作用。

GM_CSF系列靶点蛋白

粒细胞-巨噬细胞集落刺激因子(GM-CSF)通过与其受体GM-CSFR结合,可促进细胞的增殖、存活和分化,发挥多种生物学作用。但GM-CSF的异常表达会导致过度的炎症、疼痛、趋化和组织损伤,并促进其他致病细胞因子的产生。

G_CSF系列靶点蛋白

粒细胞集落刺激因子(G-CSF)主要作用于中性粒细胞系造血细胞的增殖、分化和活化。在体外刺激骨髓造血细胞的中性粒细胞前体,使之分化增殖形成成熟的粒细胞集落,又可提高中性粒细胞的吞噬能力,促进超氧化物的产生和碱性磷酸酶的合成。
