描述(Description)

The Human IL-7 R alpha/CD132 (Luc) HEK293 Reporter Cell was engineered to not only express STAT5 signaling response element, but also express the receptors full length human IL-7 R alpha (Gene ID: 3575) and CD132 (Gene ID: 3561). When stimulated with human IL-7 protein, the IL-7 R alpha/CD132 interaction drives STAT5-mediated luminescence. Neutralization of biological effect of human IL-7 protein by corresponding antibody results in a decrease in luminescence.

Co-expression analysis of human IL-7 R alpha and CD132 on Human IL-7 R alpha/CD132 (Luc) HEK293 Reporter Cell by FACS.
Cell surface staining was performed on Human IL-7 R alpha/CD132 (Luc) HEK293 Reporter Cell or negative control cell using FITC-labeled anti- IL-7 R alpha antibody and APC-labeled anti-CD132 antibody.

Passage Stability

IL-7 R alpha & IL-2 R gamma PASSAGE

Passage stability analysis by Signaling Bioassay.
The continuously growing Human IL-7 R alpha/CD132 (Luc) HEK293 Reporter Cell was stimulated with serial dilutions of human IL-7 protein. Human IL-7 protein stimulated response demonstrates passage stabilization (fold induction and EC50) across passage 8-24.

背景(Background)

Interleukin-7 receptor subunit alpha (IL7Ra) is also known as CD antigen CD127, which belongs to the type I cytokine receptor family and type 4 subfamily. IL7Ra /CD127 contains 1 fibronectin type-III domain. IL7Ra /CD127 is expressed on various cell types, including naive and memory T cells and many others. IL7Ra /CD127 is receptor for interleukin-7 and also acts as a receptor for thymic stromal lymphopoietin (TSLP). The IL7 receptor is a heterodimer of IL7R and IL2RG. The TSLP receptor is a heterodimer of CRLF2 and IL7R.
IL2RG/CD132 is an important signaling component of many interleukin receptors, including those of interleukin -2, -4, -7 and -21, and is thus referred to as the common gamma chain. Mutations in this gene cause X-linked severe combined immunodeficiency (XSCID) and X-linked combined immunodeficiency (XCID), a less severe immunodeficiency disorder.
