Bone Morphogenetic Protein 7 (BMP7) is a member of growth factor of the TGF-beta superfamily that plays essential roles in many developmental processes, including embryogenesis, hematopoiesis, neurogenesis and skeletal morphogenesis. BMP-7 Initiates the canonical BMP signaling cascade by associating with type I receptor ACVR1 and type II receptor ACVR2A. BMP-7 can signal through non-canonical pathways such as P38 MAP kinase signaling cascade that promotes brown adipocyte differentiation through activation of target genes, including members of the SOX family of transcription factors.

Influence of 2-Hydroxyisocaproic Acid on Growth Factor Release From Dentin in Regenerative Endodontic Treatments. A Laboratory Study

Abstract: This study aimed to assess the effect of 2-hydroxyisocaproic acid (HICA) on the TGF-β1, IGF-1, BMP-7, and VEGF-A release from root canal dentin, compared to calcium hydroxide (CH) and double antibiotic paste (DAP) in regenerative endodontic treatments (RET). Forty-eight single-canal teeth were prepared to mimic immature teeth and treated according to the RET protocol. Teeth were divided into three experimental (HICA, CH, and DAP) and one control (non-dressed) group. TGF-β1, BMP-7, IGF-1 and VEGF-A levels were measured using an ELISA kit on days 1 and 7. Significantly higher TGF-β1 release was found in the HICA and CH groups than in the other groups (p < 0.05). While the levels of growth factor release were notably higher on day 7 in the HICA group (p < 0.05), the values remained consistent across both time intervals within the control group. HICA proved to be effective as an intracanal medicament in enhancing the release of growth factors.© 2025 Australian Society of Endodontology Inc.
Mother to Child Non-Vascularized Fibula Transfer in Congenital Pseudoarthrosis of Tibia

Abstract: Congenital pseudoarthrosis tibia (CPT) is a relatively rare disease, characterized by anterolateral bowing of the tibia, non-union, and limb length discrepancy. Various surgical treatments have been described in literature for its management with differing favorable outcomes.In this report, we present a case of 3-year-old child with CPT of Crawford type IV, with associated fibular dysplasia. Maternal fibula was harvested and used as a bone graft and was stabilized by intra-medullary fixation. Complete union was achieved at 1 year after the primary surgery. No re-fractures were seen in a follow-up period of 2 years.Using maternal fibula as an alternative to use as a bone graft in the management of congenital pseudoarthrosis tibia may prove beneficial. Moreover, it is cheaper and readily available and needs less surgical expertise when compared to its alternatives such as use of bone morphogenetic protein 7, allogenic cadaveric grafting, or use of vascularized fibular graft.Copyright: © Indian Orthopaedic Research Group.
Bone Morphogenetic Protein 7 Improves Wound Healing in Diabetes by Decreasing Inflammation and Promoting M2 Macrophage Polarization

Abstract: Diabetic foot ulcers (DFUs) are a devastating complication of diabetes, presenting limited treatment success rates due to their complex pathophysiology. Bone morphogenetic protein 7 (BMP7) confers tissue protective and regenerative functions, but its potential role in diabetic wound healing is unknown. The aim of this study was to investigate the effects of topical BMP7 treatment in wound healing using a streptozotocin-induced diabetic mouse model. The expression of markers of wound healing progression were detected using RT-PCR or immunohistochemistry. Overall, BMP7 improved wound closure, as well as maturation of granulation tissue and collagen deposition, as evidenced by hematoxylin and eosin and Masson's trichrome histological analysis. The expression of inflammatory markers (IL-6, TNF-α) and matrix metalloproteinase-9 were decreased in BMP7-treated wounds, together with the number of pro-inflammatory M1 macrophages and T lymphocytes. The number of anti-inflammatory M2 macrophages was increased in BMP7-treated wounds. Moreover, BMP7 decreased oxidative stress and increased Ki67+ cells and CD31+ cells, indicating induced proliferation and angiogenesis in the wound bed compared to the control wounds. Finally, BMP7 activated the ERK pathway and suppressed the p38 pathway in diabetic wounds. Together, our data suggest that BMP7 enhanced skin wound healing in diabetes by decreasing local inflammation and oxidative stress, which promoted a regenerative environment for collagen deposition, wound maturation, cell proliferation, and angiogenesis. These findings underline BMP7 as a potential therapeutic agent for the treatment of skin wounds in diabetes.
